本講義は原野幸治博士(物質・材料研究機構,主幹研究員)が担当する.近年の計測技術の発展により,分子一つ一つ,またその集合体を原子レベルの解像度で可視化できる時代が到来した.従来の分析手法では多数の分子の時間および空間的な平均としての情報から分子構造の情報を得ていたが,これらの顕微手法は混合物に含まれる一つ一つの分子の「個性」を調べながら,その統計情報としてその物質の性質を研究することが可能となった.本講義では,分子を観察するための顕微鏡技術を中心に,最新の分子構造解析手法についてその基礎を学び,様々な研究例を紹介しながら,今後の分子科学にこれらの分析手法がどのように寄与できるかを議論する.
This lecture is given by Dr. Koji Harano (Principal Researcher, National Institute for Materials Science). Recent advances in measurement technology have brought us an era in which we can visualize individual molecules and their aggregates with atomic-level resolution. Conventional analytical methods obtain information on molecular structures from the temporal and spatial averages of many molecules, but these microscopic techniques make it possible to study the properties of materials as statistical information while examining the "individuality" of each molecule in a mixture. In this lecture, we will learn the basics of the latest molecular structure analysis techniques, focusing on microscopic techniques for observing molecules, and discuss how these analytical techniques can contribute to molecular science in the future, while introducing various research examples.
あらゆる生命現象は、階層的な生体構造中においてタンパク質、DNA、RNA、脂質、糖などの様々な生体分子が動的に相互作用することで発現しています。生命現象を分子レベルで理解するために、構造生物学による生体高分子の構造情報、生化学や生物物理学的手法による機能およびダイナミクス解析、次世代シークエンス技術とバイオインフォーマティクスによる遺伝情報解析、先端的なイメージング手法によるin vivo観察などが複合的に使われます。本授業では現代の生命科学を推進する重要な研究手法の基礎と具体的な研究例を学びます。
Various biological phenomena are expressed as the results of dynamic interactions of biomolecules such as proteins, DNA, RNA, lipids and sugars in multilevel structures of living systems. To understand these phenomena at the molecular level, various technologies and methodologies are used including the structural analyses of biopolymers, the biochemical and biophysical analyses of protein functions and dynamics, the genetic information analyses using bioinformatics and next generation sequencing, and the in vivo observations based on the advanced imaging techniques. In this seminar, students study basics and applications of the important research methods that propel the current molecular life sciences.
細胞生物学を分子レベルで研究するために必要な基本的な技術(遺伝子増幅、プラスミドの構築、遺伝子組換え体の作成、遺伝子組換えタンパク質の発現・精製など)を理解し、実践する。また、生体分子の細胞内における機能を解析するために必要な細胞生物学的な解析方法(遺伝子導入、蛍光顕微鏡による観察など)を理解し、実施する。
In this training course, students will understand and perform the fundamental techniques that are used to study cell and molecular biology, such as PCR, transformation, plasmid preparation, SDS-PAGE, and fluorescence microscopy.
地球上には多様な生物が存在するが,これらが共通して持っている基本的なしくみがある.生命現象を支える分子と細胞の基本的なはたらきについて理解することを目指す.もやもやした「生命の神秘」の曖昧さから解放し,「単純な原理に基づいて組み立てられている複雑さ」が実現している,もう一つの「神秘」に案内したい.なお,高校生物の知識は要求しない.むしろ化学の知識を使うことが多い,これもできるだけ基礎的なところから解説する.

細胞生物学的実験を通じて、現代の生物学の基盤をつくっている細胞生物学を体感してもらう。具体的には、細胞内小器官(オルガネラ)を蛍光顕微鏡により観察し、オルガネラの機能や細胞内の分子物流システムについて理解を深める。
The aim of the course is to introduce basic methods in cell and molecular biology. The course includes practical exercises in modern experimental techniques and instruments that include live cell fluorescent microscopy.
近年、分子レベルにおける生命科学の解明がすすみ、様々な化学反応が生命を維持するのに働くことが詳細にわかってきた。これらの化学反応の多くは非常に微小な変化であるが、遺伝子発現や様々なタンパク質間相互作用の制御に重要な働きを持つ。本特別講義では「生命科学における化学反応」に焦点を絞り、生体成分である核酸、蛋白の相互作用や機能を分子レベルで理解することを目的とする。特に、水素結合や疎水相互作用など、弱い相互作用の特徴とその共同効果ならびにエントロピーとエンタルピーの補償則についても詳細に説明する。
In recent years, the understanding of life science at the molecular level has advanced and it has become clear in detail that various chemical reactions work to maintain life. Although many of these chemical reactions only induce changes at the molecular level, they play an important role in controlling gene expression and various protein-protein interactions. In this special lecture, we will focus on "Chemical Reactions in Life Science" and aim to understand the interaction and function of biological components such as nucleic acids and proteins at the molecular level. In particular, the properties and synergetic effects of weak interactions, such as hydrogen bonding and hydrophobic interactions, will be explained in detail. The rules of entropy and enthalpy compensation are also described.
