分子細胞生物学の基礎実験
Basic experiment of cell and molecular biology
細胞生物学を分子レベルで研究するために必要な基本的な技術（遺伝子増幅、プラスミドの構築、遺伝子組換え体の作成、遺伝子組換えタンパク質の発現・精製など）を理解し、実践する。また、生体分子の細胞内における機能を解析するために必要な細胞生物学的な解析方法（遺伝子導入、蛍光顕微鏡による観察など）を理解し、実施する。
In this training course, students will understand and perform the fundamental techniques that are used to study cell and molecular biology, such as PCR, transformation, plasmid preparation, SDS-PAGE, and fluorescence microscopy.
細胞生物学を分子レベルで研究するために必要な基本的技術の原理を理解し、実践する。
The aim of this course is that students learn about the basic techniques for cell and molecular biology.
約６日間の日程で、以下の操作について実施する。
・遺伝子の増幅
・プラスミドへの遺伝子組み込みと大腸菌への遺伝子導入
・大腸菌からのプラスミドの精製
・組換えタンパク質の発現と精製
・ＳＤＳ-ポリアクリルアミドゲル電気泳動（PAGE）
・培養細胞への遺伝子導入
・培養細胞の蛍光顕微鏡観察法
・PCR
・Plasmid construction and transformation of bacteria
・Preparation of plasmids
・Expression and purification of recombinant proteins from bacteria
・SDS-PAGE
・Plasmid transfection to mammalian cultured cells
・Fluorescent observation of mammalian cultured cells
出席（３０％）とレポート（７０％）によって評価する。ただし、出席日数が全体の2/3に満たない場合や、指定した締切日までにレポートが提出されない場合、原則として単位を認定しないので注意すること。
Evaluation is performed based on attendance (30%) and submitted assignments (70%).
実習マニュアルを配付する。
Manuals are handed out on the first day.
参考書などを用いて実習の内容を予め予習してくることを強く推奨する。
Students are required to collect information and topics related to the content of the training using textbooks and appropriate websites.
