生体を構成する主成分であるタンパク質はアミノ酸が連結し折り畳まれた三次構造を有し、生物学的に重要な機能を発揮する際には、局所的もしくは広域的な構造変化を起こす。その動的な構造変化を原子レベルで捉え理解することは、生命現象の深い理解に重要である。本授業では、生体分子の機能と反応及びその立体構造について理解を深め、X線結晶構造解析を中心とした構造解析手法とその応用について習得する。
Proteins are a main component of living organisms with a tertiary structure in which linked amino acids are folded. When proteins exhibit biologically important functions, they undergo local or global structural changes. Dynamic protein structure information at the atomic level leads to a better understanding of life phenomena. In this course, students will gain more insight into functions, reactions, and structures of biomolecules. Students will also learn about protein structure analysis including mainly X-ray crystallography and their applications.
あらゆる生命現象は、階層的な生体構造中においてタンパク質、DNA、RNA、脂質、糖などの様々な生体分子が動的に相互作用することで発現しています。生命現象を分子レベルで理解するために、構造生物学による生体高分子の構造情報、生化学や生物物理学的手法による機能およびダイナミクス解析、次世代シークエンス技術とバイオインフォーマティクスによる遺伝情報解析、先端的なイメージング手法によるin vivo観察などが複合的に使われます。本授業では現代の生命科学を推進する重要な研究手法の基礎と具体的な研究例を学びます。
Various biological phenomena are expressed as the results of dynamic interactions of biomolecules such as proteins, DNA, RNA, lipids and sugars in multilevel structures of living systems. To understand these phenomena at the molecular level, various technologies and methodologies are used including the structural analyses of biopolymers, the biochemical and biophysical analyses of protein functions and dynamics, the genetic information analyses using bioinformatics and next generation sequencing, and the in vivo observations based on the advanced imaging techniques. In this seminar, students study basics and applications of the important research methods that propel the current molecular life sciences.

1.目的
バイオテクノロジーは近年著しい進歩を遂げている。この授業ではそれを支える生物化学の基礎、とりわけ、生体構成物質の構造と機能の基礎知識について学ぶ。

これまで学んできた有機化学や物理化学の知識を基にして、生体機能発現と密接に関連するアミノ酸やタンパク質の構造、生体触媒としての酵素の構造と機能、およびその反応速度論、補酵素の種類と役割などについて学ぶ。

1. Purpose
By solving exercises in applied chemistry, organic chemistry, and biotechnology, students gain a further understanding of the knowledge obtained from related lectures.
2. Overview
Based on the knowledge obtained through lectures related to organic chemistry and biotechnology, students practice fundamental concepts and applications of methodologies for structure determination and functional analysis of organic molecules and biomolecules. Based on the knowledge obtained through lectures related to surface chemistry and material physical chemistry, students practice inorganic chemistry-related structural chemistry and physical property evaluation problems.
3. Learning Goals
Students will interpret spectra of compounds and biomolecules obtained from various measuring instruments, which are essential in chemical research, and obtain information on molecular structures and functions. Students will understand the basic concepts of inorganic chemistry and apply it.
生命情報の流れのセントラルドグマ(DNA→RNA→タンパク質)に沿って、配列情報であるゲノム情報から実際の機能の担い手であるタンパク質へと、生命情報がデジタル(文字列情報)からアナログ(タンパク質立体構造情報)へと伝わっていく過程について順を追って解説して行く。遺伝子レベルでは、遺伝子構造とプロモーター解析を通じて文字列情報の解析手法について解説し、RNA レベルでは遺伝子の発現量情報解析を通じて数値データの解析を説明する。また、タンパク質レベルでは機能発現に重要な立体構造データ(3次元構造データ)の扱いについて解説する。アルゴリズムそのものよりは、生命情報の理解と言う応用面において情報科学が果たす役割に重点を置いて解説する。
生体分子の構造形成において重要な役割を果たす分子間相互作用について学ぶ。さらに、生体分子の構造、細胞の状態解析のための主要な手法である電子吸収・蛍光・赤外吸収・ラマン散乱・円偏光二色性・ESR・NMR・X線回折について、それらの原理を学び、生体分子の構造および細胞内状態の解析などへの応用を理解する。本科目は1年次に開講される「物理化学1」および全学教育科目「化学A」の内容を踏まえて行われる。
This course provides students with basic knowledge of intermolecular interactions forming structures of biomolecules and the principles and concepts of a variety of spectroscopic methods for measuring biomolecular structures. The spectroscopic methods treated are X-ray diffraction, UV-Vis absorption, fluorescence, circular dichroism, IR, Raman, NMR, and ESR. Students are recommended to have finished "Physical Chemistry 1" and "Chemistry-A" before taking this course.
