Course Title

Laboratory in Molecular Biology

授業の目的と概要

生命機能の分子レベルでの解明には遺伝学および分子生物学的手法が不可欠である。本実習では、モデル生物である線虫C. elegansを用いて、遺伝学・分子生物学的手法の基礎を学ぶことを目的とする。

Purpose/Abstract

Genetic and molecular biological techniques are essential for analyzing biological phenomena at the molecular level. In this course, basics of genetic and molecular biological experiments will be performed using the nematode C. elegans as a model system.

学習の到達目標

線虫C. elegansをモデル系として、野生型と突然変異体、形質転換体の解析を行うことにより、遺伝学と分子生物学の基礎を学ぶ。

Goal

To learn the basics of genetics and molecular biology by using the nematode C. elegans as a model system.

線虫の分子遺伝学
実験1 線虫C.elegansの基礎的取り扱いと野生型観察
実験2 変異体の表現型観察と連鎖解析
実験3 微分干渉顕微鏡による線虫胚発生過程の観察
実験4 DAPI染色を用いた個体内の種々の細胞と核の観察
実験5 RNA干渉（RNAi）法による遺伝子機能阻害と表現型観察
実験6 線虫C.elegansの走性実験
実験7 線虫近縁種の観察

Molecular Genetics of the nematode C. elegans

1) Basic handling of C. elegans
2) Observation of mutant phenotypes and linkage analysis
3) Observation of C. elegans embryogenesis with differential interference contrast (DIC) microscopy
4) Observation of morphology of various cells and nuclei by DAPI staining and GFP-fusion proteins
5) Gene knock-down by RNAi and phenotype analysis
6) Chemotaxis assay of C. elegans
7) Observation of related nematode species
